IMO the writers first we're going to make it some type of one dies another is made. Later the changed the plan to genetics. So when we get to Josh we don't know if Josh will become a Grimm like his dad because of the genetic factor. If the writers had left it the other way than Josh would have become a Grimm.
It wasn't guaranteed that he would become a Grimm. He didn't have a Monroe and Rosalee to woge for him at the time so I don't see how he would believe her. Marie would've had to force some Wesen to woge in front of Nick unless he happened to see one fully woged like Hank did with Monroe and Wu did with the Aswang. Who knows how those two would've turned out if not for Nick explaining thing to them although he should've come clean to Wu long before he did. Wu should still be bitter about that whole episode.
I don't even know how Marie knew Nick was showing signs when she showed up. Nick accepted what she was saying only because his abilities were manifesting. He could've turned out to be another Josh and thought just thought of her as crazy ol' Aunt Marie.
Taking that angle Marie thought that maybe because she wasn't Nick's mom he wouldn't become a Grimm until his mom died.
