Trubel said she knows of 2 other Grimm's will she use this to help Nick with the hunt for the keys? If the were 7 Grimms to start with could it take 7 to end it?
Hmm, now that's interesting. Way back in S2 Monroe told Nick about the map very likely depicting a part of the Schwarzwald. He also spoke of the key being meant to open part of a 'ward lock' (Nothing to do with Harry Potter!)
Now there are a few different definitions of a ward lock. But, the one I think is important here is the one that says it is a type of lock made of several different barriers that have to be opened by several different keys.
Last year my old buddy Starfury and I discussed this here on the Forum. What we thought was that each key opened one, and only one of seven barriers! And God help you if you used key#4 in barrier #5!!
But what if there is an extra twist? Each key has to used by a direct descendant of each of the 7 Grimm knights? Each key is magically clued to each knights DNA. So, if Truble tries to use Nick's key - Boom!! Or something of the sort.

DNA maybe we can assume Nick is one and if Josh is now a Grimm we can assume he is one now take the Grimm Trubel knows with a key also assume he is also linked to the first Grimm's, but how would you find the other 4 Grimm from the keys the royals have? With Josh that would give us 5 Grimms find 2 more and you have your seven. Another question do all Grimm's know of the keys? I would go with questions only a Grimm could answer but what would be those questions? This falls to those who like puzzles, what questions could only a Grimm answer?
After doing the mystical cell they had Adalind in. I can only imagine what hides the keys. We know Nick finds another key, but how does he get the four that the Royals have. Will they have Nick use the three pieces of the map, and use monroes knowledge to fill in the rest. Will the third key be found by Trubel, tracing information about her parents. You could then have Nick with his family key. Trubel with her family key. Josh now having become a Grimm with his family key. They could go to the black forest together. HW and Eve follows them with plans to take what ever the keys hide. So HW can use it against the up rising. Now that Sean family is almost gone that royal key could be up for grabs.

I've got a out of box thought what if they find the 3 key where whatever is behind the lock door? Going to @wfmyers1207 point on "Synchronicity" Nick is the one to find the hidden items, now that him and Adalind are together this now completes this part of the puzzle for the keys to be found. Also out of the box what if the door also hides the item which the Grimm's move when the royals started getting the keys.

because of "Synchronicity" i was thinking the kids where needed. Maybe it will be a Grimm and Hexenbiest working together.

You know never thought of the kids. I your idea how would you work it? I had thought about maybe a hexen but dropped that because I just didn't see Grimm's of era working with hexen, but maybe that was the trick because no one would.
